It's important to first make sure your current employer is aware that you are searching for a new position. This should not come as a surprise if you are approaching graduation. I would say to them that you are starting to apply for various positions, and there have been a couple that really excite you, but they are asking for a referral. Then ask your manager if they would feel comfortable providing you with one. Most people will be happy to do this if you've been a good worker, shown a strong work ethic, have been on time to work, etc. I would just say the key here is making your employer aware that you've started looking for a new position.
